By searching a database for sequences similar to C. elegans Wdr23, Choe
et al. (2009) identified 2 splice variants of human DCAF11, which they
called WDR23. The deduced 546- and 520-amino acid WDR23 isoforms differ
only in the presence or absence of a 26-amino acid insert near the N
terminus. Both isoforms contain 7 WD40 repeats and a conserved DW box
motif that overlaps WD40 repeat 5.
GENE FUNCTION
The transcription factor Skn1 (POU2F3; 607394) protects C. elegans from
stress and promotes longevity. Choe et al. (2009) showed that Wdr23
interacted with Skn1 and Ddb1 (600045) in C. elegans. Loss of Wdr23
function caused accumulation of Skn1 in intestinal nuclei, elevation of
Skn1 protein levels, constitutive transcription of detoxification genes,
and increased longevity and stress resistance. Wdr23 interacted with the
Ddb1-Cul4 (see CUL4A; 603137) complex and presumably targeted Skn1 for
proteasomal degradation. Choe et al. (2009) concluded that Wdr23 is a
negative regulator of Skn1 in C. elegans.
MAPPING
Hartz (2010) mapped the DCAF11 gene to chromosome 14q11.2 based on an
alignment of the DCAF11 sequence (GenBank GENBANK AF130070) with the
genomic sequence (GRCh37).
