CLONING
Using N-terminally truncated RANBPM (RANBP9; 603854) as bait in a yeast
2-hybrid screen of a human lymphocyte cDNA library, followed by PCR,
Umeda et al. (2003) cloned C20ORF11, which they called TWA1. The deduced
228-amino acid protein has a calculated molecular mass of 30 kD. TWA1
has an N-terminal LIS1 (PAFAH1B1; 601545) homology (LISH)/C-terminal to
LISH (CTLH) motif, which is found in proteins involved in microtubule
dynamics, cell migration, nucleokinesis, and chromosome segregation. It
also has a C-terminal domain similar to a C-terminal domain in RANBPM.
TWA1 shares 99.5% amino acid identity with mouse Twa1 and is well
conserved among mammals. Database analysis revealed TWA1 homologs in
fly, plant, and yeast. Northern blot analysis showed ubiquitous Twa1
expression in mouse tissues. Western blot analysis detected a 30-kD TWA1
protein in Hela cells and several mammalian cell lines.
GENE FUNCTION
Using coimmunoprecipitation analysis, Umeda et al. (2003) showed that
endogenous Ranbpm and Twa1 interacted in COS-7 cells.
Immunoprecipitation and gel-filtration analyses demonstrated that both
proteins interacted with GST-fused human muskelin (MKLN1; 605623) in a
670-kD protein complex in transfected COS-7 cells.
MAPPING
The International Radiation Hybrid Mapping Consortium mapped the
C20ORF11 gene to chromosome 20 (TMAP RH78524).
