CLONING
Using the differential display technique, Kocher et al. (1996)
read moreidentified a epithelial-specific gene, originally designated DD96, which
was markedly upregulated in various human carcinomas originating from
the kidney, colon, lung, and breast, as compared with normal epithelial
cell populations. By Northern blot analysis and in situ hybridization,
Kocher et al. (1995) found that, in normal tissues, MAP17 is expressed
at significant levels only in a single epithelial cell population, the
proximal tubular epithelial cells of the kidney. By Western blot
analysis, Kocher et al. (1996) determined that the DD96 protein product
is a 17-kD membrane-associated protein, which they redesignated MAP17.
By immunofluorescence studies, they localized MAP17 to the apical brush
border membrane. In permeabilized cultured cell lines, MAP17 was
associated with cell-cell membrane contact areas, indicating that MAP17
is an intracellular protein associated with the cytoplasmic aspect of
the plasma membrane. Transfection of a full-length wildtype MAP17 cDNA
clone into a colon carcinoma cell line markedly decreased cell
proliferation in vitro and tumor growth in vivo.
GENE FUNCTION
Using the yeast 2-hybrid system, Kocher et al. (1998) identified PDZK1
(603831) as a protein interacting with MAP17. By in situ hybridization,
they showed that both proteins are upregulated in human carcinomas
arising in kidney, lung, colon, and breast.
MAPPING
Gottgens et al. (2000, 2001) demonstrated that the MAP17 gene lies
downstream of the stem cell leukemia gene SCL (187040) in human, mouse,
and chicken genomes; Gottgens et al. (2002) showed that it is located in
a comparable position in zebrafish as well. Human MAP17 maps to
chromosome 1p33.

CLONING
Using the differential display technique, Kocher et al. (1996)
read moreidentified a epithelial-specific gene, originally designated DD96, which
was markedly upregulated in various human carcinomas originating from
the kidney, colon, lung, and breast, as compared with normal epithelial
cell populations. By Northern blot analysis and in situ hybridization,
Kocher et al. (1995) found that, in normal tissues, MAP17 is expressed
at significant levels only in a single epithelial cell population, the
proximal tubular epithelial cells of the kidney. By Western blot
analysis, Kocher et al. (1996) determined that the DD96 protein product
is a 17-kD membrane-associated protein, which they redesignated MAP17.
By immunofluorescence studies, they localized MAP17 to the apical brush
border membrane. In permeabilized cultured cell lines, MAP17 was
associated with cell-cell membrane contact areas, indicating that MAP17
is an intracellular protein associated with the cytoplasmic aspect of
the plasma membrane. Transfection of a full-length wildtype MAP17 cDNA
clone into a colon carcinoma cell line markedly decreased cell
proliferation in vitro and tumor growth in vivo.
GENE FUNCTION
Using the yeast 2-hybrid system, Kocher et al. (1998) identified PDZK1
(603831) as a protein interacting with MAP17. By in situ hybridization,
they showed that both proteins are upregulated in human carcinomas
arising in kidney, lung, colon, and breast.
MAPPING
Gottgens et al. (2000, 2001) demonstrated that the MAP17 gene lies
downstream of the stem cell leukemia gene SCL (187040) in human, mouse,
and chicken genomes; Gottgens et al. (2002) showed that it is located in
a comparable position in zebrafish as well. Human MAP17 maps to
chromosome 1p33.
