DESCRIPTION
The vacuolar proton-ATPase (V-ATPase) translocates protons into
intracellular organelles or across the plasma membrane of specialized
cells. The catalytic domain consists of a hexamer of 3 A subunits and 3
B subunits, plus accessory subunits C, D (607028), and E (108746).
CLONING
Van Hille et al. (1993) cloned subunit C from a human osteoclastoma cDNA
library with probes developed by PCR from the bovine cDNA sequence
(Nelson et al., 1990). The deduced 382-amino acid human ATP6C protein
has a calculated molecular mass of 41,941 Da and shows 99% sequence
identity with bovine ATP6C. Northern blot analysis detected ubiquitous
and comparable expression of a 1.9-kb transcript and a fainter doublet
of 6.0-7.0 kb.
MAPPING
The International Radiation Hybrid Mapping Consortium mapped the
ATP6V1C1 gene to chromosome 8 (TMAP WI-9077).
