CLONING
Using a human autoimmune serum against the nucleolar organizer region
(NOR), Bolivar et al. (1999) isolated a hamster cDNA that they called
NOA36. They used this cDNA as a probe to select human ZNF330 cDNAs from
a HeLa cell library. The human ZNF330 gene encodes a 320-amino acid
protein with a predicted molecular mass of 36.3 kD. The ZNF330 protein
contains a putative nuclear localization signal, 9 conserved CXXC zinc
finger motifs, and an acidic C terminus. EST analysis identified ZNF330
homologs in hamster, mouse, Drosophila, and nematode. Northern blot
analysis showed expression of a 1.8-kb ZNF330 transcript in all tissues
examined, with highest expression seen in heart and skeletal muscle.
GENE FUNCTION
By Western blot analysis using recombinant polypeptides and by
immunofluorescence studies, Bolivar et al. (1999) confirmed that ZNF330
is recognized by anti-NOR serum. Further immunofluorescence studies in
mammalian cells demonstrated that ZNF330 localizes primarily to the
nucleolus during interphase and to centromeres during mitosis. They also
showed that after inhibition of transcription by actinomycin-D
treatment, ZNF330 remains associated with the nucleolus, but no longer
colocalizes with the nucleolar factor UBTF (600673).
Using RNAse treatment, Bolivar et al. (2001) showed that the nucleolar
association of ZNF330 is RNA-dependent.
GENE STRUCTURE
Bolivar et al. (2001) showed that the ZNF330 gene contains 10 exons and
spans 16 kb.
MAPPING
Using FISH and genomic database analyses, Bolivar et al. (2001) mapped
the ZNF330 gene to 4q31.1-q31.2.
