CLONING
Palmiter and Findley (1995) cloned rat Znt1. The 5-prime UTR is GC rich,
and the deduced protein contains 507 amino acids. Znt1 has 6
membrane-spanning domains and intracellular N and C termini. Northern
blot analysis detected low expression of 2.4- and 7.0-kb transcripts in
all mouse tissues examined. Znt1 localized to the plasma membrane of
transfected baby hamster kidney cells.
By EST database analysis, Seve et al. (2004) found evidence of moderate
SLC30A1 expression in B cells and leukocytes, with lower expression in
bone, brain, breast, cervix, colon, head, neck, kidney, lung, ovary,
pancreas, parathyroid, placenta, prostate, stomach, and uterus.
GENE FUNCTION
Segal et al. (2004) found mouse Znt1 modulated zinc permeation through
the L-type calcium channel following expression in human embryonic
kidney cells and rat neuronal precursor cells. Znt1 downregulated not
only Zn(2+) influx, but also Ca(2+) influx, thereby protecting cells
from the effects of excessive cation permeation.
GENE STRUCTURE
Palmiter and Findley (1995) determined that mouse Znt1 contains 2 coding
exons. The 5-prime flanking region is part of a GC island, and the
promoter region has a TATA-like motif and 2 potential metal response
elements.
Seve et al. (2004) stated that the human SLC30A1 gene contains 2 exons.
MAPPING
Seve et al. (2004) stated that the SLC30A1 gene maps to chromosome
1q32.3.
